Calculates DNA melting temperature from GC content using Tm = 81.5 + 16.6 log[Na⁺] + 0.41(%GC) − 675/n (Marmur-Doty formula). 1% more GC increases Tm by ~0.41 °C — PCR primer design uses Tm to set annealing temperature 5 °C below the lower primer Tm.
